So we are excited to see new studies about proteins in shiitake mushrooms called lentiavidins. You might recognize the "avidins" part of this name from research on egg whites. Avidins are egg-white proteins that can bind together with biotin. In the case of shiitake, researchers have added the designation "lenti"—from the shiitake genus Lentinula—to come up with the new protein name, "lentiavidins."
Lentinan, an immunostimulant derived from shiitakes, has been used to treat cancer, AIDS, diabetes, chronic fatigue syndrome, fibrocystic breast disease, and other conditions with impressive results.

Shiitake has interested researchers due to its medicinal properties, and several biologically active compounds have been isolated and purified from the mushrooms, mycelia and aqueous extract (Chang, 1999; Ishikawa et al., 2001; Hassegawa et al., 2005). These compounds exhibit antitumoral, antifungal, antibacterial, hypoglycemic and antioxidant properties (Maeda et al., 1998; Ngai and Ng, 2003; Ishikawa et al., 2001; Yang et al., 2002; Kitzberger et al., 2007) that can have one or more therapeutic effects and can act synergistically (Pascholati et al., 1998; Wasser and Weis, 1999). Among these compounds, some of the best characterized are lentinan (Chihara et al., 1969; Minato et al., 1999), KS-2, (Fujii et al., 1978), lentinacin (2(R), 3(R)-dihidrixi-4-(9 adenil)- butyric acid), known as eritadenine (Shimada et al., 2003a; Shimada et al., 2003b, Enman et al., 2008), and fruit body protein -(FBP), which inhibits the tobacco mosaic virus (Kobayashi et al., 1987. Lentinamycin (octa-2, 3-diene-5, 7-diyne-1-ol) was isolated and purified from a liquid culture of L. edodes (Bew et al., 1966). This compound inhibits the growth of Trichoderma sp (Tokimoto et al., 1987) and Gram-positive bacteria (Komemushi et al., 1996). Wow - so as my shiitake log mycelium fight off the Trichoderma invasion - they are increasing their lentinamycin levels and therefore also their Lentinan levels! This is why Lentinan is higher in log grown mushrooms.... in the forest, grown in the wild.

Effects of Management on the Yield and High-Molecular-Weight Polysaccharide Content of Shiitake (Lentinulaedodes) Mushrooms
High-molecular-weight polysaccharides (HMWP) including lentinan in shiitake may promote human health. This study was conducted to determine if management protocols influence the HMWP of shiitake mushrooms. Results indicate that measuring the total carbohydrate content of water-extractable, ethanol-insoluble polysaccharides was a simple way to estimate HMWP. Results also indicate that log-grown shiitake contained more HMWP than did substrate-grown shiitake.

Shiitake mushrooms contain lentinan, a polysaccharide that has numerous medicinal benefits. The objective of this research was to determine the impact of various rates of chipped and sawmill bark added to the substrate on lentinan found in shiitake mushrooms. Crude polysaccharide extract (CPE) and lentinan (LTN) content was significantly different among shiitake mushrooms grown on various artificial substrates containing different percentages of sawdust and chipped bark. The addition of chipped bark content from 0% to 50% increased the CPE content, but an increase in chipped bark content from 50% to 100% decreased the CPE and LTN content. CPE content in mushrooms was greater when equal proportions of sawdust and bark were used. A 100% heartwood treatment had the least CPE content.

Only the diet containing 5% of dried L. edodes consistently resulted in lower viable counts (ca. 1 – 2 log10 CFU) of total bacteria, E. coli, streptococci and lactic acid bacteria, and luminal and mucosal effects agreed very well. With this diet, acetate and butyrate concentrations in the distal jejunum were doubled, which is favourable in view of the trophic effect on enterocytes and colonocytes.

The lentinan contents in the Lentinus edodes fruit body during storage were examined by ELISA method using anti-lentinan antibodies. The lentinan content (12.8 mg.g(-)(1) dw) before storage decreased to 3.7 mg.g(-)(1) dw over 7 days at 20 degrees C. However, it only slightly decreased at 1 degrees C and only decreased to 9.3 mg. g(-)(1) dw at 5 degrees C. Glucanase activity, which seems to be associated with lentinan degradation, increased more during storage of L. edodes at 20 degrees C than it did at lower temperatures. In addition, only glucose was detected as a degraded product from lentinan by the glucanase. This suggested that this enzyme would fit the profile of an exo-type glucanase. Also, polyphenol oxidase activity, known as an index of freshness reduction in the mushroom, increased approximately 2.7-fold (to 61.5 units.mg(-)(1)) over 7 days during storage at 20 degrees C. However, its activity changed little during storage at lower temperatures.
